CRISSY – Automated Salt Pre-Screening and Polymorph Screening

CRISSY

CRISSY is the ideal tool for salt pre-screening and polymorph screening for extensive crystallisation studies, as it allows the screening of a range of solvents in the eluotropic series. The preparation and crystallisation process runs on a specially designed workbench which automates the necessary steps using different solvents, concentrations, temperature gradients, agitation, pH measurement, filtration and controlled evaporation. It is also possible to carry out an array of recrystallisation on the salt crystals.

A specially designed reactor block presents 96 crystals directly to the XRPD system, thereby eliminating crystal damage. The top surface of the reactor base is ideal for XRPD-detection. Cross contamination caused by electrostatic forces from moving the powders, is minimized as the block is grounded and the individual reactor cavities are separated from each other.

CRISSY makes polymorph screening easy. Automating the crystallisation is not only time-saving but it increases the quantity of salts being crystallised and minimises the risk of destroying the crystal structure when presenting it to the x-ray. The reliability of the instrument means that the process can be run in an uninterrupted manner and the controlled liquid handling, heating and cooling cycles ensure reproducible and consistent results.



ProCrys – Automated Liquid and Powder Handling Workstation

ProCrys

ProCrys is an automated sample preparation workstation for protein crystallisation. It combines a liquid handling platform for the fast preparation of the buffer solutions with a low-volume dispenser for the preparation of the crystallisation plates and a plate sealer.

Buffer solutions are prepared by filling stock solutions into source (deepwell) plates. The software calculates the required volumes for a given target concentration and also optimises the entire dispensing process to save time. All the user has to do is to create a file, e.g. in Excel, which contains the barcodes of the source plates and the required concentrations for the stock solutions.

A low volume dispenser with a separate protein channel is used to prepare the crystallisation plates. The buffer solutions are transferred from the deepwell plate to the crystallisation plate in one step with a special 96-well dispensing head. The protein is added through the seperate protein channel via non-contact dispensing.


The plates are sealed on the integrated plate sealer which seals the deepwell plates as well as the crystallisation plates.

A special version, the ProCrys Meso, is also available for membrane proteins in lipidic mesophases. The ProCrys Meso is a new liquid handling platform for the preparation of 96 in-meso (or cubic phase) proteins. It offers accurate and precise dispensing of the protein and the screening solution and also can be used to prepare your own screening solutions on the workbench instead of using the more expensive commercial kits. An integrated module ensures the ideal humidity conditions for the process. Clever tools have been incorporated to set up a run more easily, including precise and reproducible dispensing of the mesophase.



SuSY – Automated Pre-Formulation & Solubility Screening Workstation

SuSY

SuSy is a new automatic workbench which enables you to execute a workflow to test a large number of compounds in parallel. The reactors are based on the microtitre format with either 24, 48 or 96 reactors. The APIs are precisely weighed into the reactors. Based on the weight the required amount of aqueous and organic solutions, as well as the viscous components, are added. For thorough mixing the sample rack is moved to temperature controlled vortexers or magnetic stirrers. Automatic sealing of the reactors prevents evaporation of the samples. Tools for pH-measurement and adjustment are also available for integration.

For analysing the dissolution status of the solid compounds, an image is taken using a camera pick-up tool and analysed. Based on the analysis, additional treatment such as sonication is applied and then a further image is taken to confirm the dissolution.

Once all the compounds have been dissolved, daughter plates are prepared and sealed on the integrated plate sealer for further use. Evaporation, filtration and HPLC/LC-MS preparation can be integrated and samples for analysis can be taken at any time.
